چکیده

BACKGROUND Renal cell carcinoma (RCC) is characterized by its resistance to radiotherapy and/or chemotherapy. On the other hand, it is an immunogenic tumor - it is able to stimulate antitumor responses. A prognostic significance of HLA-G expression by neoplastic cells in RCC is not well characterized; significance HLA-E expression in RCC is not characterized at all. METHODS In our study, we evaluated the expression of HLA-G and HLA-E specific mRNA transcripts produced by neoplastic cells in 38 cases of RCC and in 10 samples of normal kidney parenchyma. The results were statistically correlated with various clinico-pathological parameters. RESULTS We confirmed that HLA-G is downregulated in normal kidney tissue; if it is up-regulated in RCC, then it is connected to worse prognosis. On the other hand, HLA-E mRNA transcripts were present in both normal kidney tissue and RCC and their increasing concentrations counterintuitively carried better prognosis, more favorable pT stage and lower nuclear Fuhrmann's grade. CONCLUSION Considering the fact that there is known aberrant activation of HLA-G and HLA-E expression by interferons, identification of HLA-G and HLA-E status could contribute to better selection of RCC patients who could possibly benefit from more tailored neoadjuvant biological/immunological therapy. Thus, these molecules could represent useful prognostic biomarkers in RCC, and the expression of both these molecules in RCC deserves further study. Prognostic Value of HLA-G in Malignant Liver and Pancreas Lesions

Background: Human leukocyte antigen (HLA)-G is a nonclassical HLA class I molecule with modulatory effects on NK and T cells. Because HLA-G expression is frequently detected in different solid tumors, it may be involved in tumor immune evasion. Objective: This study was designed to elucidate the prognostic value of HLA-G in hepatocellular carcinoma (HCC) and pancreatic adenocarcinoma (PADC). Th...

Background: HLA-G, as a tolerogenic molecule, expressed in cytotrophoblast cells and playing an important role in the suppression of the immune response in maternal decidua. Today Interaction of the extracellular domains of HLA-G protein with cell receptors of immune system, including CD8, LILRB1, LILRB2 and killer cell immunoglobulin-like receptor KIR2DL4, was well known. The Effect of Interferon Beta in HLA-G Expression on Monocyte in Diabetes Type1

Abstract Background and objectives: Dendritic Cells are the most important of antigen presenting cells with an effective role in immune tolerance. This study, aims to clarify the role of IFN- β in induction on dendritic cells derived by monocyte in diabetes type1 to evaluate the T cells response to beta cell specific antigenic molecule. Material and Methods: In this research, peripheral bl...

Natural Killer Cell Cytotoxicity Against SKOV3 after HLA-G Downregulation by shRNA

Background: HLA-G is a nonclassical HLA class I molecule which, when elevated in tumor cells, is one of the main factors involved in tumor evasion of immune responses including NK and T cells. Objective: To evaluate the effect of HLA-G downregulation on NK cell cytotoxicity in tumor cell lines. Methods: The expression level of HLA-G was measured by real-time PCR and flowcytometry after transfec...

HLA-G-specific microRNAs a novel approach for targeting tumors

Immune inhibitory molecules on immune cells as well as on tumor cells appear to play a key role in modulating the immunogenicity of tumors and the efficacy of immunotherapies. One mediator is represented by the nonclassical HLA-G antigen, which is often overexpressed in human tumors when compared to corresponding normal tissues thereby inhibiting both innate as well as adaptive immune responses...
